Small tissue culturing success

I started trying to tissue culture mid January. Besides tomato seed these are the only things i've been successful with so far. Dionaea Muscipula leaf and seed. The leaf explant was put in vitro on February 13th. More than a month later and there are lil shoots everywhere.

Congrats! The hard part, at least for me, is transitioning from TC to au nateurelle.
 
I tried that with some tomatoes I had in vitro. Didn't go so well for me either. I'm thinking about buying some kind of coating spray, maybe a wax spray, to keep them from dying off ex vitro. Guess i'll see when the time comes. Thanks for stopping by jim!
 
Nice pics dionae! I am getting my first TC kit anytime now :) I was wondering what you were using for a surface sterilant? Also, is your growth medium custom mixed?

Household bleach, peroxide and alcohol. I don't have any certain technique. Still trying to find what I like the most. Last few explants I used bleach/water mix only, no rinse. Probably use a different process next time. I'm still a noob and trying to find what works best for me:).

is your growth medium custom mixed?
No, my mix is simple.

1/3 MS w/ vitamins
.5-1ml BAP or Kinetin
1ml PPM
30g sugar

I add agar by the container so it varies and use the microwave to sterilize. I bought a pressure cooker but have had so lil contamination with the microwave i've never used it. I hope you have lots of success and good luck!

Thanks NY Plant Nerd. Wish me luck on getting them to callus. Cant seem to find too many people that have successfully TCed explants and if they have theyre keeping protocols a secret:p. I'm going to keep at it until I finally get a piece TCed. Thanks for stopping by!
